iPhone-imaged and cell-powered electrophoresis titration chip for the alkaline phosphatase assay in serum by the moving reaction boundary

Abstract: As a vital enzyme, alkaline phosphatase (ALP) has great clinical significance in diagnoses of bone or liver cancer, bone metastases, rickets, and extrahepatic biliary obstruction. However, there is still no really portable chip for the ALP assay in blood. Herein, a simple electrophoresis titration (ET) model was developed for ALP detection via a moving reaction boundary (MRB). In the model, ALP catalyzed the dephosphorylation of a 4-methylumbelliferyl phosphate disodium salt (4-MUP) substrate in the cathode we… Show more

“…ALP activity could be quantified by measuring the distance of MRB. The mobile phone can be used for data collection and analysis ( Figure 6B) [132]. Recently, Wang et al also used a smart phone, to determine anthrax based on color change, using Eu(III) functionalized carbon dots (CDs-Eu) as a fluorescent probe, where DPA (an anthrax biomarker) can interact with CDs-Eu, resulting in a color change.…”

“…When compared to SECM imaging, electrochemical imaging, using LRC-EC systems is appropriate for continuous imaging of biomaterials since LRC-EC makes it possible to achieve rapid image acquisition. Cao et al (2018) created a simple electrophoresis titration (ET) device for ALP detection through moving reaction boundary (MRB) [171]. In this model, ALP sped up the dephosphorylation of the 4-methylumbelliferyl phosphate disodium salt substrate in the cathode and 4-methylumbelliferone (4-MU) with blue fluorescence and negative charge under UV excitation.…”

“…In recent decades, a variety of strategies using colorimetry [2,[7][8][9][10][11][12][13][14], fluorescence [15][16][17][18][19], surface-enhanced Raman scattering [20], and electrochemistry [21,22] have been established for detecting ACP. Compared with other traditional detection methods, colorimetric assays have advantages such as fast response, simple operation, real-time visualization, and low cost.…”
