PKC delta activation increases neonatal rat retinal cells survival in vitro: Involvement of neurotrophins and M1 muscarinic receptors

Abstract: Protein kinase C (PKC) is a family of serine/threonine kinases related to several phenomena as cell proliferation, differentiation and survival. Our previous data demonstrated that treatment of axotomized neonatal rat retinal cell cultures for 48 h with phorbol 12-myristate 13-acetate (PMA), a PKC activator, increases retinal ganglion cells (RGCs) survival. Moreover, this treatment decreases M1 receptors (M1R) and modulates BDNF levels. The aim of this work was to assess the possible involvement of neurotrophi… Show more

“…PRKCD overexpression reversed the effect of TRIM69 overexpression on BDNF expression. A previous study showed that PRKCD activation upregulated BDNF expression in retinal ganglion cells [37]. These results indicate that TRIM69 negatively regulates BDNF expression in GC cells via PRKCD.…”

“…PRKCD is a member of the protein kinase C family of serine-and threonine-speci c protein kinases [36,37] and is involved in the progression of GC [38,39]. For example, PRKCD overexpression enhances the metastasis, chemoresistance, and stem cell-like characteristics of GC cells [38].…”

“…Therefore, over the last few years, the therapeutic involvement of cholinesterase inhibitors as part of neuroprotective therapy has gained relevance. 81,87,88 Similarly, some experimental studies have demonstrated that the activity of Ach precursors such as citicoline, improves retinal function, restoring the activity of the mitochondrial membrane of RGC, increasing the expression of neural growth factors, and improving cell survival, which confirms that there is a close correlation between retinal function and cholinergic activity [89][90][91] .…”

“… 42 , 43 When comparing cPKCγ +/+ +MD mice and cPKCγ −/ − +MD mice, the amplitudes of the a-waves and b-waves in the right eye were not statistically different, indicating that cPKCγ did not affect light transmission of retina. This phenomena might be due to the fact that other subtypes of PKC play an important role in retina function, such as aPKCζ, 44 , 45 nPKCδ, 44 , 46 and cPKCα. 47 , 48 In addition, the intraocular pressures of the MD and control eyes were 8.67 ± 0.380 mm Hg and 8.5 ± 0.365 mm Hg, respectively (n = 6 per group; t = 0.316; degrees of freedom = 10; P = 0.758).…”
