Prospective Analyses of Cytokine Mediation of Sleep and Survival in the Context of Advanced Cancer

Abstract: Sleep duration was associated with symptom burden and poorer survival and IL-2 was found to mediate the association between sleep and survival. Screening and treatment of sleep problems in patients diagnosed with cancer are warranted.

“…IL-1 plays a key role in carcinogenesis and tumor growth [ 193 ], due to its importance as a key downstream mediator of inflammation [ 194 ]. Increased levels of IL-1β in body fluids are correlated in experimental tumor models and in cancer patients with bad prognosis, carcinogenesis and invasiveness of the tumor [ 195 , 196 , 197 ].…”

“…Increased levels of IL-1β in body fluids are correlated in experimental tumor models and in cancer patients with bad prognosis, carcinogenesis and invasiveness of the tumor [ 195 , 196 , 197 ]. The key mechanisms by which IL-1β promotes tumor development are driving chronic non-resolved inflammation [ 193 ], endothelial cell activation [ 194 ], tumor angiogenesis [ 195 ] and induction of immunosuppressive cells ( Figure 3 ). Altogether, these mechanisms account for suppression of the adaptive immunity, tumor promotion and metastasis [ 193 ].…”

“…165,166 For example, insomnia associated with cancer has been linked to decreased quality of life, 165 impaired immune function, 167,168 cancer-related fatigue, 169 and may have implications for tumor progression [170][171][172] and greater mortality risk. 173 These secondary medical conditions (eg, sleep disruption) can aggravate inflammatory processes and stimulate the release of central cytokines, which act to alter neuronal plasticity and brain functioning, affecting peripheral biobehavioral processes such as the activation of the HPA axis and the SNS, resulting in the production of high levels of cortisol. Thus, stress management interventions might help to reduce, mitigate, or prevent chemobrain syndrome indirectly by mitigating the risk of and effects of comorbidity or multimorbidity.…”
