C. elegans MRP-5 Exports Vitamin B12 from Mother to Offspring to Support Embryonic Development

Abstract: SUMMARY Vitamin B12 functions as a cofactor for methionine synthase to produce the anabolic methyl donor S-adenosylmethionine (SAM) and for methylmalonyl-CoA mutase to catabolize the short-chain fatty acid propionate. In the nematode Caenorhabditis elegans, maternally supplied vitamin B12 is required for the development of offspring. However, the mechanism for exporting vitamin B12 from the mother to the offspring is not yet known. Here, we use RNAi of more than 200 transporters with a vitamin B12-sensor trans… Show more

“…Animals were washed in 0.9% saline until the solution was clear and then twice more (3–6 times total). Metabolites were extracted and analyzed as described previously ( Na et al, 2018 ). Briefly, 50 µL of a semi-soft pellet of animals was transferred to a 2 mL FastPrep tube (MP Biomedicals) and flash frozen with liquid nitrogen.…”

“…After settling, supernatant was transferred to glass vials (Sigma) and dried by speed-vac overnight. MeOX-MSTFA derivatized samples were analyzed on an Agilent 7890B/5977B single quadrupole GC-MS equipped with an HP-5ms Ultra Inert capillary column (30 m × 0.25 mm×0.25 μm) using the same method as described ( Na et al, 2018 ).…”

“…In addition to folate, all the other vitamins of the B complex were also reported to be essential for normal growth and development of C. briggsae in axenic medium: riboflavin, thiamine, pyridoxine, niacinamide, pantothenic acid, biotin and cobalamin [55, 184–186]. Among these, cobalamin (vitamin B12) was most extensively studied in the C. elegans model system [30, 187189]. This vitamin has the unique property of being synthesised solely by archaea and bacteria [190].…”

“…C. elegans is a bacterivore, making it a highly suitable "simple" model organism to study vitamin B12 biology Zhang et al, 2017). For instance, we have previously found that bacterial diets high in vitamin B12 accelerate C. elegans development and change metabolic gene expression Na et al, 2018;Watson et al, 2014). One set of genes repressed by vitamin B12 comprise an alternative propionate breakdown pathway, or propionate shunt (Watson et al, 2016) (Figure 1).…”
