Transcript Profiling and Gene Identification Involved in the Ethylene Signal Transduction Pathways of Creeping Bentgrass (Agrostis stolonifera) during ISR Response Induced by Butanediol

Abstract: Creeping bentgrass (Agrostis stolonifera) is the preferred green lawn grass, with excellent turf characteristics but poor disease resistance. At present, the mechanisms of disease resistance in creeping bentgrass are poorly understood, especially the ethylene signal transduction pathway under the induced systemic resistance (ISR) response. In this study, butanediol (BDO), as a new type of disease-resistance compound, was applied to creeping bentgrass seedlings to induce the ISR response. Then, we measured ethy… Show more

“…To refer to chemicals which trigger IR, a plethora of terms has been introduced, mostly not accompanied by a clear definition, characterization or thorough differentiation from others: 'resistance activator' [33], 'plant (defense) activator' [34][35][36], 'synthetic inducer of defense responses' [37], 'defense elicitor' [38], 'inducer of plant immunity' [39], '(plant) resistance inducer' [40,41], 'disease resistance compound' [42], 'elicitor' [43,44], 'inducer' [45], 'SAR inducer' [17], 'plant strengthener' [46,47], 'priming-inducing chemical' [26], 'priming agent' [48], et cetera. As these terms tacitly might be associated with specific underlying mechanisms -'priming agents' may be thought of as mainly leading to primed defense responses, which can be conceived as opposed to the mode-of-actions of 'plant defense activators' -the parallel use of these terms can be confusing to newcomers in the field.…”

“…Creeping bentgrass ( Agrostis stolonifera ), is one of the major cool-season grass species, which is widely used as turf and forage grass in temperate regions. Previous studies have been conducted in creeping bentgrass under either drought or heat stress condition, in order to understand physiological, molecular, transcriptomic and proteomic responses related to drought or heat tolerance 21 25 . However, no information regarding to the common regulation patterns for creeping bentgrass under either drought or heat stress condition is available.…”

“…Using mRNA as a template, first-strand cDNA was synthesized using random primers and reverse transcriptase. Second-strand cDNA was synthesized using RNase H and DNA polymerase I (Takara Bio, Inc.) ( 25 ). Subsequently, the cDNA was purified, and subjected to end-repair, poly(A) addition, sequencing, adapter connecting and fragment size selection.…”
