Investigation of the effects of thymoquinone on erythrocyte deformability in sepsis treatment which created by cecal perforation in rat

Abstract: AIM: We aimed to study the effects of thymoquinone on erythrocyte deformability in an experimental model of sepsis given before or after the initiation of the sepsis model. METHOD: The animals were grouped as (n = 6) control, nigella sativa, sepsis, sepsis group with administration of nigella sativa before sepsis development and sepsis group with nigella sativa administration after sepsis development. Cecal ligation and puncture model (CLP) was used to induce sepsis in the animals. The thymoquinone was given 1… Show more

“…Leptocytes and codocytes were noted in hepatic insufficiency, especially in dogs and cats with portocaval shunts [ 27 , 29 ]. Sepsis leads to decreased red blood cell deformability in rats and mice [ 30 , 31 ] and altered erythrocyte morphology, with the formation of echinocytes, sphero-echinocytes, and spherostomatocytes [ 32 ]. These changes are thought to either be a combined result of oxygen free radicals and toxin action, or depletion of ATP in cells [ 32 ].…”

“…In order to create a sepsis model, cecal ligation and puncture (CLP) procedure were performed on 44 rats as previously described (16,17). 8 rats were excluded from the study because they died.…”

“…Moreover, thymoquinone was found to prevent sequels of the multiple organ failure syndrome of sepsis via regulating the production of NO and its inflammatory sequela, and adjustment of vascular responsiveness (Alkharfy et al, 2015). Thymoquinone restored the erythrocyte deformability observed in CLP-induced sepsis in rats, showing more effectiveness when applied before the establishment of the sepsis (Bostanci et al, 2018). Ozer et al (2017) found that thymoquinone intraperitoneally injected in rats at 1 mg/kg exhibited protective effects on mesenteric perfusion, contractile function of aorta as well as anti-inflammatory and antioxidative effects.…”
