Summary Objectives Our study aimed to update the seroprevalence and factors associated with anti‐dengue virus (DENV) antibody positivity among blood donors and to discuss their implications for blood supply. Background Questions on the potential transmission of DENV by transfusion increased after the documentation of the risk of transmission of the West Nile virus. This risk was estimated after transfusion of DENV RNA‐positive blood units of up to 37.5%. In Burkina Faso, very few studies on DENV in blood donors have been conducted. As a result, there were no reliable data on DENV to allow the implementation of appropriate measures to control the risk of transmission of the dengue virus by blood transfusion. Methods We conducted a 4‐week cross‐sectional study from December 4 to 30, 2016. Blood donors of both genders, aged 18‐60 years, accepted for blood donation after medical selection were consecutively enrolled. Results Our study included a total of 1007 blood donors, in which donors living in urban areas represented 78.2%. The mean age was 26.1 ± 8.1 years. After adjustment in a multiple regression logistic model, the odds of having IgG anti‐DENV increased as age increased. The odds of DENV was 53% lower in rural areas (OR = 0.47; P = .000) compared to urban settings and 42% lower in mobile sites (OR = 0.58; P = .03) compared to fixed ones. Conclusion Our study provides new and useful insights for future research on the risk of TT‐DENV throughout blood transfusion.

IntroductionDengue is the most important mosquito-borne diseases worldwide but was considered scarce in West-Central Africa. During the last decade, dengue outbreaks have increasingly been reported in urban foci in this region suggesting major epidemiological changes. However, in Central Africa where both vectors, Aedes aegypti and Aedes albopictus are well established, the role of each species in dengue transmission remains poorly investigated. Methodology/Principal findingsField-collected strains of Ae. aegypti and Ae. albopictus from different ecological settings in Central Africa were experimentally challenged with dengue 2 virus (DENV-2). Mosquitoes were analysed at 14-and 21-days post-infection. Analysis provide evidence that both Ae. aegypti and Ae. albopictus in Central Africa were able to transmit dengue virus with Ae. aegypti exhibiting a higher transmission rate. Unexpectedly, two Ae. aegypti populations from Bé noué and Maroua, in northern Cameroon, were not able to transmit DENV-2. Conclusions/SignificanceWe conclude that both Ae. aegypti and Ae. albopictus are susceptible to DENV-2 and may intervene as active dengue vectors. These findings highlight the urgent need to plan a vector surveillance program and control methods against dengue vectors in Central Africa in order to prevent future outbreaks. Globally, 390 million people are at risk of dengue infection and over the past 50 years, the virus incidence increased thirty‐fold. In Senegal, an unprecedented occurrence of outbreaks and sporadic cases have been noticed since 2017. In October 2018, an outbreak of Dengue virus 2 (DENV‐2) was reported in the north of Senegal affecting multiple areas including Saint‐Louis, Richard Toll, and Rosso which are located at the border with Mauritania. Of these 173 blood specimen samples collected from patients, 27 were positive for dengue by quantitative reverse transcription PCR (qRT‐PCR), and eight were serologically confirmed to be positive for DENV immunoglobulin M (IgM). Serotyping using qRT‐PCR reveals that isolates were positive for DENV‐2. A subset of DENV‐2 positive samples was selected and subjected to whole‐genome sequencing followed by phylogenetic analysis. Analysis of six nearly complete genome sequences revealed that the isolates belong to the cosmopolitan genotype and are closely related to the Mauritanian strains detected between 2017 and 2018 and those detected in many West African countries such as Burkina Faso or Cote d'Ivoire. Our results suggest a transboundary circulation of the DENV‐2 cosmopolitan genotype between Senegal and Mauritania and call for a need for coordinated surveillance of arboviruses between these two countries. Interestingly, a high level of homology between West African isolates highlights endemicity and calls for the set‐up of subregional viral genomic surveillance which will lead to a better understanding of viral dynamics, transmission, and spread across Africa.
