Galectin-3 impacts Cryptococcus neoformans infection through direct antifungal effects

Abstract: Cryptococcus neoformans is an encapsulated fungal pathogen that causes cryptococcosis, which is a major opportunistic infection in immunosuppressed individuals. Mammalian β-galactoside-binding protein Galectin-3 (Gal-3) modulates the host innate and adaptive immunity, and plays significant roles during microbial infections including some fungal diseases. Here we show that this protein plays a role also in C. neoformans infection. We find augmented Gal-3 serum levels in human and experimental infections, as wel… Show more

“…More recently, it was also shown to play a role in C. neoformans infection, as galectin-3-deficient mice were found to be more susceptible to cryptococcosis than wild-type mice. In addition, galectin-3 was able to inhibit fungal growth and exert a direct lytic effect on C. neoformans extracellular vesicles protecting the host from potential fungal virulence factors carried in these vesicles [49]. …”

“…Fungal EVs carry several virulence factors and other important molecules, contributing to fungal pathogenicity and host immunomodulation (914). Since EVs plays significant roles in the host-pathogen relationship, the vesicular stability is important to ensure suitable delivery of their cargo into host cells (13, 15).…”

“…These interactions commonly affect the microorganism pathogenesis, the host immune response or the success of intracellular parasitism (1619). Recently, we have reported that Galectin-3 (Gal-3), a β-galactoside-binding animal lectin, plays significant roles in cryptococcal infection (13). Gal-3 interferes the C. neoformans infection, inhibiting C. neoformans growth and promoting vesicle disruption (13).…”
