Downregulation of the Sonic Hedgehog/Gli pathway transcriptional target Neogenin-1 is associated with basal cell carcinoma aggressiveness

Abstract: Basal Cell Carcinoma (BCC) is one of the most diagnosed cancers worldwide. It develops due to an unrestrained Sonic Hedgehog (SHH) signaling activity in basal cells of the skin. Certain subtypes of BCC are more aggressive than others, although the molecular basis of this phenomenon remains unknown. We have previously reported that Neogenin-1 (NEO1) is a downstream target gene of the SHH/GLI pathway in neural tissue. Given that SHH participates in epidermal homeostasis, here we analyzed the epidermal expression… Show more

“…Furthermore, when specifying the subtypes of BCC analyzed, we found the increase of strong levels of Shh immunoexpression in aggressive variants – infiltrative, mixed, and micronodular. These results agree with the data demonstrated by Casas et al [ 60 ]. By contrast, among the other subtypes of BCC, previously considered as aggressive variants, we found upregulation of Shh paralleled by downregulation of α -SMA immunoexpression in the superficial subtype of the tumor assumed to be nonaggressive.…”

These results are in agreement with the data demonstrated by Casas and colleagues(Casas et al, 2017).By contrast, among the other subtypes of BCC, previously considered as aggressive variants, we found upregulation of SHh paralleled by downregulation of α-SMA expression in the superficial subtype of the tumour assumed to be nonaggressive. This leads us to propose that SHh participates in normal and affected epidermal homeostasis, but the molecular pathway of the signalling is not completely understood.…”

“…Paraffin-embedded patient samples of NB were deparaffinated and rehydrated as described in [ 21 ]. IHC assays were proceeded by incubating the slides with primary antibodies anti-NEO1 (1:50, sc-15,337, Santa Cruz biotechnologies), anti-NTN1 (1:40, AF6419, sheep, R&D systems) and anti-PCNA (1: 100, 13–3900, mouse, Invitrogen) antibodies in 2.5% horse serum (from the Vestatin kit).…”
