A matrix-assisted laser desorption/ionization time of flight mass spectrometry reference database for the identification of Histoplasma capsulatum

Abstract: The isolation of the pathogenic fungus Histoplasma capsulatum from cultures together with the visualization of typical intracellular yeast in tissues are the gold standard methods for diagnosis of histoplasmosis. However, cultures are time-consuming, require level 3 containment and experienced personnel, and usually call for an additional confirmation test. Matrix-Assisted Laser Desorption Ionization Time of Flight Mass Spectrometry (MALDI-ToF MS) has been established as a suitable tool for microbial identific… Show more

“…Isolation of the fungus in culture is considered the gold standard for diagnosis. H. capsulatum can grow in mycological media of common use in routine laboratories, and current MALDI-TOF systems can provide a presumptive identification (Panda et al, 2015;Rizzato et al, 2015;Rychert et al, 2018;Valero et al, 2018). Cultures, however, have well-known limitations, i.e., slowness, suboptimal sensitivity, and requirement of BSL3 facilities to manipulate them (Kauffman, 2009).…”

“…Alternatively, single-stranded DNA probes could also be used to detect the fungus in cultures (AccuProbe, Hologic, CA, USA), however the procedure is technically complex and time consuming [16]. In last years, matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-ToF MS) has started to be developed for the identification of H. capsulatum strains [17][18][19]. Although reports are still scarce, results in this area are promising since the system is able for recognizing different varieties (var.…”
