Arctiin regulates collagen type 1α chain 1 mRNA expression in human dermal fibroblasts via the miR-378b-SIRT6 axis

Abstract: Arctiin, a lignin isolated from Arctium lappa, exhibits a variety of biological effects, including anti‑viral, anti‑inflammatory, and anti‑proliferative actions, in mammalian cells. In a previous study, arctiin was demonstrated to induce procollagen type I synthesis and exhibited protective effects against ultraviolet B (UVB) radiation in normal human dermal fibroblasts (nHDFs). However, the underlying molecular mechanism of arctiin‑mediated collagen synthesis remains unknown. In the present study, the mechani… Show more

“…A previous study has shown that miR-378b is expressed in normal human dermal fibroblasts, and circulating miR-378b is a sensitive biomarker for photo-aging (Joo et al, 2017). It was also found to protect dermal fibroblasts from UVB-inhibited collagen I by targeting SIRT6 (Kim et al, 2017). However, the role of miR-378b in the pathogenesis of hepatic insulin resistance remains elusive.…”

“…This subsequently represses the mRNA expression levels of a-1-type 1 collagen (COL1A1) by disrupting SIRT6 activity ( 77 , 78 ). Arctin, a lignin derived from Arctium lappa , downregulates microRNA378b, preventing UVB-induced reduction in COL1A1 and restoring SIRT6 activity ( 78 ). Recently, in vitro studies by Lee et al revealed that UV-irradiated dermal fibroblasts had lowered SIRT2 and SIRT3 mRNA and protein levels ( 79 ).…”

“…Specifically, microRNA-378b is induced in UVB-exposed human dermal fibroblasts. This subsequently represses the mRNA expression levels of a-1-type 1 collagen (COL1A1) by disrupting SIRT6 activity (77,78). Arctin, a lignin derived from Arctium lappa, downregulates microRNA378b, preventing UVB-induced reduction in COL1A1 and restoring SIRT6 activity (78) ), hydrogen peroxide (H 2 O 2 ), hydroxyl radicals (•OH), and singlet oxygen ( 1 O 2 ).…”

“…miR-128 inhibits osteoblast differentiation in osteoporosis by downregulating SIRT6 expression, thus accelerating the development of osteoporosis [ 19 ]. miR-378b represses the mRNA expression levels of COL1A1 via interference with SIRT6 in human dermal fibroblasts [ 20 , 21 ].…”
