Thymic stromal lymphopoietin contribution to the recruitment of circulating fibrocytes to the lung in a mouse model of chronic allergic asthma

Abstract: Our data suggested that TSLP might function in airway remodeling by promoting circulating fibrocyte recruitment to the lung in the mice subjected to chronic allergen exposure. These results provide a better rationale for targeting the interaction between TSLP and fibrocytes as a therapeutic approach for chronic allergic asthma.

“…We have previously demonstrated that production of TSLP was increased in the lung of asthmatic mice and TSLP level is associated with DC maturation and the extension of Th2‐biased inflammation . Our data showed that neutralization of TSLP with corresponding monoclonal antibody obviously alleviated allergen‐induced airway eosinophilic inflammation, inhibited production of Th2‐associated cytokines in the lung and subsequent airway remodeling . Individuals with one atopic disease are far more likely to develop a second disease.…”

“…As an epithelium‐derived cytokine, thymic stromal lymphopoietin (TSLP) has been suggested to be a master switch that initiates allergic inflammation in the lung and other tissues, including in those with asthma . In addition, we and others have demonstrated that TSLP was also involved in allergen‐induced airway remodeling by acting on airway structural cells, such as recruiting fibrocytes into the lung tissues, inducing airway smooth muscle cells migration, promoting fibroblast differentiation . These findings highlight the potential advantages of targeting TSLP, which may affect disease activity more broadly than the inhibition of a single downstream pathway.…”

“…Recruitment of fibrocytes into wound areas forms the basis of their functions (4). Thymic stromal lymphopoietin has been reported to function in airway remodeling by promoting CF recruitment to the lungs in mice subjected to chronic allergen exposure (7). In addition, contribution of chemokines chemokine ligand (CCL)5, CCL11 and CCL24 in the recruitment of fibrocytes to the airway in severe asthma has also been previously demonstrated (8).…”
