A resource of large-scale molecular markers for monitoring Agropyron cristatum chromatin introgression in wheat background based on transcriptome sequences

Abstract: Agropyron cristatum is a wild grass of the tribe Triticeae and serves as a gene donor for wheat improvement. However, very few markers can be used to monitor A. cristatum chromatin introgressions in wheat. Here, we reported a resource of large-scale molecular markers for tracking alien introgressions in wheat based on transcriptome sequences. By aligning A. cristatum unigenes with the Chinese Spring reference genome sequences, we designed 9602 A. cristatum expressed sequence tag-sequence-tagged site (EST-STS) … Show more

“…Our marker-based approach of identifying disomic lines was supported by GISH results. Therefore, the use of molecular markers from the genomes of important wild relatives to track their chromosomes in wheat is a reliable method to facilitate speedy expansion of diversity in wheat germplasm 31 , 35 , 36 . This approach would incentivize wheat breeders to develop cultivars with improved adaptability to keep pace with the projected global increase in wheat demand 37 .…”

“…P genome-specific markers and 6P-specific STS markers [ 30 , 32 ] were used to detect translocation lines and deletion lines. P genome-specific markers can trace the P genomic component in wheat background, which were designed through specific DNA sequences distributing the entire P genome [ 30 ].…”

“…Genomic DNA was extracted using a modified CTAB method ( Kidwell and Osborn, 1992 ; Li et al, 2017 ). A total of 82 A. cristatum chromosome 2P-specific STS markers which were developed based on the A. cristatum transcriptome sequences ( Zhang et al, 2015 , 2017 ) were mapped to 17 2P-specific bins, and the information of STS markers was described as Li et al (2017) . Of these, eight markers located on different 2P-specific bins were selected to detect the absence or presence of 2P chromosomal segments in the translocation lines and their backcross populations ( Table 4 ).…”
