Vitamin D supplementation to palliative cancer patients: protocol of a double-blind, randomised controlled trial ‘Palliative-D’

Abstract: A randomised, double-blind placebo-controlled multicentre trial has been designed. The trial will include 254 adult palliative cancer patients with 25-OHD levels <50 nmol/L and a life expectancy of more than 3 months recruited from two advanced palliative home care centres in Stockholm. Included patients will be randomly assigned to 12 weeks of treatment with cholecalciferol (vitamin D3) 4000 IU/day or placebo. The study will start in November 2017 and will finish in December 2019. The study is approved by the… Show more

“…In addition, there was a decrease in infections after three months of vitamin D supplementation as well as an increase in self-assessed quality of life. The results from this pilot study were used for the power-calculation of a future randomized, placebo-controlled, double-blind study called “Palliative-D” that will start in November 2017 and will include n = 254 palliative cancer patients [ 68 ]. The primary endpoint in that study will be “change in opioid dose” between placebo and vitamin D-treated patients.…”

“…The first study was a randomized, placebo-controlled, double-blind study on vitamin D in palliative care, “The Palliative-D study” performed during 2017–2020 [ 17 , 18 , 19 ]. Serum samples from the screening visit were used, i.e., before any study drug had been given.…”

“…It has been suggested that correction of vitamin D deficiency may have a positive effect on fatigue in palliative cancer patients [ 52 , 53 , 54 ]. This is currently investigated in a large ongoing randomized, placebo-controlled, double-blind study, “Palliative-D” [ 55 ]. The results from this study are still not published but in the baseline data from the study cohort ( n = 530) there was a significant association between low vitamin D levels and severe fatigue in men but not in women [ 56 ].…”
