Gauging and Visualizing c-di-GMP Levels in Pseudomonas aeruginosa Using Fluorescence-Based Biosensors

Abstract: Recent research has shown that the molecule c-di-GMP is an important second messenger regulating various functions in bacteria. In particular, the implication of c-di-GMP as a positive regulator of adhesion and biofilm formation has gained momentum as a highly relevant research topic, as detailed knowledge about the underlying regulatory mechanisms may enable the development of measures to control biofilms in both industrial and medical settings. Accordingly, it is in many cases of interest to measure the c-di… Show more

“…2A and B ). This indicates that P. aeruginosa bacteria in the planktonic state contain a low level of c-di-GMP, which is in agreement with previous work ( 35 ). The p lac -gfp -tagged strain showed a biofilm-like aggregation morphology at 8 h postinfection (hpi) in our mouse model of corneal infection ( Fig.…”

“…To further address whether the intracellular c-di-GMP level in P. aeruginosa is elevated during corneal infection, we used P. aeruginosa carrying a p cdrA - gfp gene fusion, which is a well-established c-di-GMP reporter strain ( 34 36 ). In the planktonic state, the c-di-GMP reporter bacteria exhibited an extremely low expression level of the p cdrA - gfp fusion (below the detection limit) compared with a P. aeruginosa strain that carries a p lac -gfp fusion and constitutively expresses the green fluorescent protein (GFP) gene gfp ( Fig.…”

“…Intracellular levels of c-di-GMP were assessed with the fluorescence-based biosensor pCdrA-gfpC (63, 64), acquired as addgene plasmid #111614; http://n2t.net/addgene:111614 ; RRID:Addgene_111614. Purified plasmids were transformed by electroporation in evolved morphotypes obtained from static cultures (65).…”

“…C-di-GMP levels were assessed with the fluorescence-based biosensor pCdrA-gfpC (53, 54). pCdrA-gfpC was constructed by Tim Tolker-Nielsen (addgene plasmid #111614; http://n2t.net/addgene:111614 ; RRID:Addgene_111614).…”
