Validity of Prostate Health Index and Percentage of [-2] Pro-Prostate-Specific Antigen as Novel Biomarkers in the Diagnosis of Prostate Cancer: Omani Tertiary Hospitals Experience

Abstract: Phi outperforms tPSA and fPSA when used alone or in combination, and appears to be more accurate than both markers in excluding prostate cancer before biopsy. Use of this biomarker helps clinicians to avoid unnecessary biopsies, particularly in patients with gray-zone tPSA level. Phi is the strongest marker that correlates proportionally with Gleason Score; therefore, it is also useful in predicting the aggressiveness of the disease. This is the first reported experience for the use of p2PSA and phi in Oman, t… Show more

“…When we used a threshold value of 1.66 for %p2PSA, 25 it had a sensitivity of 94.2% and a specificity of 62.5% with NPV of 71.4% and PPV of 91.5%. The AUC for %p2PSA is shown in Fig.…”

“…Additionally, PHI (total PSA, free‐PSA, p2PSA isoform), 4KScore (total PSA, free‐PSA, intact PSA, hK2), fusion between the transmembrane serine protease gene (TMPRSS2), and the v‐ets erythroblastosis virus E26 oncogene (ERG) (TMPRSS2‐ERG) are amongst the best known diagnostic biomarkers/scoring methods that have been developed relatively recently for PCa detection 59 . The SE and SP for these biomarkers/scoring methods range from 0.75 to 0.93, and 0.33 to 0.80, respectively 60–63 . Besides, a number of studies have assessed the potential of circulating miRNAs as early‐stage, localized prostate cancer biomarkers 64,65 .…”

“…PHI (prostate health index) is serum based biomarker it is a strongest biomarker. When compare to other biomarker PHI has high specificity and sensitivity which use to avoid unnecessary biopsies [14]. TNF-α, sTNFR1 and IL-8 was measured in the serum sample using ELSIA kit, it gives highly significant detection of prostate cancer in men compare to PSA [15].…”
