Experimental Modeling Supports a Role for MyBP-HL as a Novel Myofilament Component in Arrhythmia and Dilated Cardiomyopathy

Abstract: Rationale/Objective Cardiomyopathy and arrhythmias are under significant genetic influence. Here, we studied a family with dilated cardiomyopathy and associated conduction system disease in whom prior clinical cardiac gene panel testing was unrevealing. Methods/Results Whole genome sequencing and induced pluripotent stem cells were used to examine a family with dilated cardiomyopathy and atrial and ventricular arrhythmias. We also characterized a mouse model with heterozygous and homozygous deletion of Mybph… Show more

“…Cardiomyocyte differentiation and replating. Reprogrammed iPSCs were differentiated to cardiomyocytes according to previously described protocol (58), with the starting densities between 100,000-320,000 cells/ cm 2 . Differentiated cardiomyocytes were replated 5-7 days after beating was observed to 12 mm No.…”

“…Transthoracic echocardiography was performed prior to sacrifice at the indicated time points, as previously described ( 26 , 27 ). Briefly, the rats were anesthetized with 2.0% isoflurane via inhalation.…”

“…In vivo cardiac function was assessed by echocardiography and was conducted under anesthesia (0.8 liters/min, 1% vaporized isoflurane in 100% O 2 ) as described previously (Barefield et al, 2017). Echocardiography was performed using a Visual Sonics Vevo 2100 imaging system with a MS550D 22-55 MHz solid-state transducer (FujiFilm).…”
