Local IL‐25 contributes to Th2‐biased inflammatory profiles in nasal polyps

Abstract: Local IL-25 plays a crucial role in promoting Th2-biased inflammatory profiles in NP and may serve as a promising therapeutic target in CRSwNP patients.

“…Patients grouped into the high IL-25 expression cohort had higher CT sinus severity scores, higher nasal endoscopy scores, and higher downstream type-2 cytokine levels. 53 In another study, patients with high IL-25 levels were found to have nasal polyps which were more responsive to glucocorticoid therapy, 54 and this finding was corroborated in an in vitro nasal polyp explant model in which steroid exposure reduced IL-25 production. 23 …”

“…57 In CRSwNP patients, higher levels of IL-33 also correlated with higher CT sinus severity scores, higher nasal endoscopy scores, and higher downstream T H 2 cytokine levels. 53 One group demonstrated that a receptor of IL-33, ST2, is elevated in ethmoid mucosa in CRSwNP patients. 58 Interestingly, ILC2 cells from diseased mucosa from the same patients with CRSwNP produce more IL-13 in response to stimulation with recombinant IL-2 and IL-33, compared to ILC2s from controls.…”

“…There might be a potential for other soluble factors as markers of Th2 responses. Of specific interest is epithelium‐derived IL‐25, which in contrast to IL‐33 and TSLP was found significantly increased in nasal polyps of chronic rhinitis patients …”

“…Importantly, a skewed type 2 immune response is initiated with the involvement of cytokines IL‐25, IL‐33, TSLP and type 2 innate lymphoid cells (ILC2). These skew helper‐T‐cell maturation towards the Th2 fate, and direct B cells to induce Ig class switching to IgE, processes mediated by type 2 cytokines IL‐4, IL‐5 and IL‐13 (Figure ). This type 2 skewing persists in an individual, even after the initial response has passed and involuted.…”

“…Pro‐inflammatory cytokines modulate LRRK2 mRNA expression suggesting a negative role of LRRK2 during inflammation. Local IL‐25 promotes the production of Th2 cytokines in polyp tissues of CRSwNP and may become of interest as a potential therapeutic target . Myeloid dendritic cells (mDCs) present in nasal polyps have elevated surface expression of IL‐17RB and ST2, suggesting that mDCs might be actively involved in IL‐25‐ and IL‐33‐induced Th2‐type immune responses and eosinophilic inflammation .…”
