Evaluation of the Persistent Preventive Efficacy of 2.5 % Moxidectin/10 % Imidacloprid Spot-on (Advocate®, Advantage® Multi) in Dogs Experimentally Infected with Angiostrongylus vasorum

Abstract: A controlled, randomized and blinded doseconfirmation study was conducted to assess the persistent preventive efficacy of treatment with Advocate ® spot-on (2.5 % moxidectin / 10 % imidacloprid) in dogs four weeks before inoculation of third-stage larvae of Angiostrongylus (A.) vasorum. Twenty-four adult dogs were randomly allocated to three treatment groups. Dogs in group 1 were treated with Advocate ® spot-on at the minimum recommended dose of 0.1 mL per kg bodyweight (BW) on day 84 and inoculated with third… Show more

“…As the scenario of the distribution of canine parasites and VBDs is in continuous evolution, further large-scale studies are warranted for more comprehensive knowledge on the epizootiological risks in European territories, and to understand if and how the routine use of broad-spectrum endo-ectoparasiticides needs a refinement based on local settings. The use of ectoparasiticides and/or repellents is the most effective strategy to minimize the risk of diseases transmitted by arthropod vectors, while dog angiostrongylosis and dirofilariosis can be prevented by the monthly administration of macrocyclic lactones like milbemycin oxime and moxidectin [ 138 , 139 , 140 , 141 ]. Although cases of LOE/ML-resistant strain of heartworm are known to occur in North America, this is not the case in Europe [ 142 , 143 ].…”

“…For dogs recovering from A. vasorum infection with concurrent hemostatic dysfunction, the use of prophylactic treatment is recommended by the authors following successful treatment of the infection to prevent immediate re-infection. The spot-on formulation of imidacloprid/moxidectin, milbemycin oxime or moxidectin as tablets have all been approved for prevention of A. vasorum infection [13,14,[89][90][91][92][93][94][95] (Table 3). Besides these cases, prophylaxis may also be recommended in dogs with a particular high risk of infection (e.g., dogs living in highly endemic areas, dogs with a high-risk behavior such as scavenging, coprophagia, or slug/snail ingestion, dogs undergoing planned surgery, or dogs younger than 1 1 /2 years of age, as dogs this young are commonly infected) [5].…”

“…Continuation of the daily oral sildenafil (1.7 mg/kg BID) therapy was recommended until the next re-check in 2 months, and the dog was discharged. At the same time, monthly preventive moxidectin spot-on was advised for life-long use [21].…”
