Immunological properties and protective efficacy of a single mycobacterial antigen displayed on polyhydroxybutyrate beads

Abstract: SummaryIn 2015, there were an estimated 10.4 million new tuberculosis (TB) cases and 1.4 million deaths worldwide. Bacille Calmette–Guérin (BCG), an attenuated strain of Mycobacterium bovis, is the vaccine available against TB, but it is insufficient for global TB control. This study evaluated the immunogenicity of the Mycobacterium tuberculosis antigen Rv1626 in mice while assessing the effect of co‐delivering either Cpe30 (immunostimulatory peptide), CS.T3378–395 (promiscuous T helper epitope) or flagellin (… Show more

“…2B). The polyester synthase PhaC remained anchored on the surface after particle formation and isolation (29,39). Protein profile analysis by SDS-PAGE showed that the full-length PhaC-TB antigen fusions were overproduced because the MWs of fusion proteins corresponded to their theoretical MWs (Fig.…”

“…Nevertheless, the construction of bioactive PHA systems by protein immobilization has been investigated to a limited degree at the laboratory scale and is still a nascent technology that needs scaling up for industrial applications (43). Very efficient covalent methods for PHA functionalization-based translational fusions with PhaC from Ralstonia eutropha have been described in detail by Rehm's group, leading to a variety of biotechnological and biomedical applications (3,44,45). However, in the case of noncovalent methods like those based in phasins, several key technical issues, such as control of PHA-protein association/dissociation and granule size, still remain unresolved.…”

“…Moreover, IL17A, IFNγ, and IL2 are known to be important for protection against TB; however, their production postvaccination does not always correlate tightly with enhanced protective immunity. [32b,37] Future animal studies including challenge with the pathogen will investigate whether the induced immune responses prevent infection by M. tuberculosis .…”
