Vertical bone regeneration using rhBMP-2 and VEGF

Abstract: BackgroundSufficient vertical and lateral bone supply and a competent osteogenic healing process are prerequisities for the successful osseointegration of dental implants in the alveolar bone. Several techniques including autologous bone grafts and guided bone regeneration are applied to improve quality and quantity of bone at the implantation site. Depending on the amount of lacking bone one- or two-stage procedures are required. Vertical bone augmentation has proven to be a challenge particularly in terms of… Show more

“…In addition, VEGF is comprehensively expressed by cranial neural crest cells and plays multiple roles in regulating cell proliferation, vascularization, and bone formation, including endochondral ossification as well as intramembranous ossification. In addition, VEGF is widely known to induce angiogenesis 63,64 . In the process of angiogenesis, a variety of factors are involved.…”

“… Multiple factors influence the expression of bone morphogenetic proteins, including the material used as the carrier, the porosity of the implant surface, and dosage . Bone morphogenetic proteins may also be combined with other growth factors such as beta‐transcription factor proteins (TCP), vascular endothelial growth factor, and platelet‐derived growth factor‐BB to produce new bone formation . The main advantage of bone morphogenetic proteins consists of removing the need to harvest bone from a secondary donor site, thus reducing the potential for comorbidities associated with a secondary harvest .…”

“…Finally, the combination of rhBMP‐2 with other growth factors, such as BMP‐7, TGF‐β, PDGF, VEGF and bFGF, has also been suggested to enhance BIC values in canine and minipig circumferential peri‐implant defects (Luo, Zhang, Shi, Cheng & Zhang, ; Meraw, Reeve, Lohse & Sioussat, ; Sun, Wang, Zheng, Fan & Gu, ; Wang, Zheng, Zhao, Liu & Gao, ) and dehiscence‐type defects (Thoma, Cha, Sapata, Jung & Husler, ). In minipigs, enhanced vertical peri‐implant bone regeneration was also shown when adding rhBMP‐2 and VEGF to a disc‐shaped collagenous scaffold (Schorn, Sproll, Ommerborn, Naujoks & Kubler, ).…”
