Mechanistic Role of Thioredoxin 2 in Heart Failure

Abstract: Thioredoxin 2 (Trx2) is a pivotal mitochondrial protein that regulates redox signaling. The mitochondrial Trx2 is expressed ubiquitously, but it is found at the highest levels in metabolically active tissues like the heart. Global gene knockout of Trx2 results in embryonic lethality, likely due to the increased cellular oxidative stress. Moreover, mice with cardiac-specific Trx2 deletion develop spontaneous dilated cardiomyopathy (DCM), correlating with increased apoptosis stress kinase-1 (ASK1) signaling and … Show more

“…Following stimulation, ASK1 dissociates from Trx and is activated by subsequent modifications. 32 In our study, excessive oxidative stress occurred 4 weeks after MI, whereas ZBTB20 overexpression decreased ROS production and preserved the level of anti-oxidants, leading to inactivation of ASK1. In addition, ZBTB20 also decreased inflammatory response post-MI and reduced production and release of TNFα, leading to inactivation of ASK1/JNK pathway.…”

“…Myocardial cell apoptosis is central to the pathogenesis of HF. 39 Therefore, the effect of tolvaptan/valsartan combination on the apoptosis of heart and kidney tissues in Dox-treated mice was evaluated by TUNEL assay. As indicated in Figure 5 A,B, increased number of TUNEL-positive cells were detected in heart and kidney tissues of mice in the model group.…”

“…GSSG is then recycled by GSH reductase (GR) 9 . Recent studies have established an important role of the Trx antioxidant system in HF 10,11 . GSH has also been implicated in cardiovascular diseases, however, only recently have the enzymes of the GSH synthesis and salvage pathways been implicated in HF [12][13][14][15][16][17][18] .…”
