Modulation of caveolins, integrins and plasma membrane repair proteins in anthracycline-induced heart failure in rabbits

Abstract: Anthracyclines are chemotherapeutic drugs known to induce heart failure in a dose-dependent manner. Mechanisms involved in anthracycline cardiotoxicity are an area of relevant investigation. Caveolins bind, organize and regulate receptors and signaling molecules within cell membranes. Caveolin-3 (Cav-3), integrins and related membrane repair proteins can function as cardioprotective proteins. Expression of these proteins in anthracycline-induced heart failure has not been evaluated. We tested the hypothesis th… Show more

“…In our results, Cav-3 expression was increased 1.6-fold in DDC-fed TG neg hearts compared with chow-fed TG neg hearts. This result is consistent with previous reports that structural caveolae and Cav-3 protein were both increased in a rabbit model of daunorubicin cardiotoxicity (11). Therefore, the increase of Cav-3 in the hearts under liver cirrhosis might be a compensatory increase, and further studies for the protective mechanism of Cav-3 OE are needed.…”

“…The chow-fed Cav-3 OE hearts showed approximately three times higher Cav-3 protein expression than chow-fed TG neg hearts (P Ͻ 0.001). Cav-3 expression was increased 1.6-fold in DDC-fed TG neg hearts compared with chow-fed TG neg hearts (P ϭ 0.041), which is similar to our previous reports of increased Cav-3 expression in daunorubicin-treated rabbit hearts (11). There were no differences in fibrosis of the heart between the four groups (Fig.…”

“…Given that rabbit models of human heart disease have similarities to human physiology and are practical alternatives to larger mammals (Pogwizd and Bers, 2008), we used four groups of blood plasma obtained from previous studies in rabbit heart failure Models (Ichikawa et al, 2017;McKirnan et al, 2019). Brie y, in the MI experiment, New Zealand White rabbits underwent thoracotomy wherein a cryo-probe cooled with liquid N 2 was used to induce transmural MI by freezing anterior-lateral left ventricular (LV) free wall for 3 min.…”

“…Препараты вводят внутримышечно, реже подкожно [2-4, 6, 17-21]. Некоторые авторы применяют ингаляционный масочный наркоз с использованием 1,0-1,5% изофлурана (галогенизированный анестетик) -в комбинации с мидазоламом (2 мг/ кг) [13,22]. Возможна также эндотрахеальная интубация и ингаляционный наркоз 1,8% изофлураном после вводной анестезии кетамином (10 мг/ кг) и мидазоламом (0,1 мг/кг) [3].…”
