Bisindolylmaleimide alkaloid BMA-155Cl induces autophagy and apoptosis in human hepatocarcinoma HepG-2 cells through the NF-κB p65 pathway

Abstract: Bisindolylmaleimides, a series of derivatives of a PKC inhibitor staurosporine, exhibit potential as anti-cancer drugs and have received considerable attention in clinical trials. This study aims to investigate the effects of a bisindolylmaleimide alkaloid 155Cl (BMA-155Cl) with a novel structure on autophagy and apoptosis in human hepatocarcinoma HepG-2 cells in vitro and in vivo. The cell poliferation was assessed with a MTT assay. Autophagy was evaluated by MDC staining and TEM analysis. Apoptosis was inves… Show more

“…Despite the vast amount of information about modulations of PKC function in cellular models, the relevance of individual PKC isozymes in the progression of human cancer is still a matter of controversy. PKC signaling controls the several signaling pathways in the cell [32][33][34][35]. Although PKC inhibitors used in this study affect several pathways in different cellular process, they are able to specifically inhibit their targeted PKC isoform activity.…”

“…After the cells were washed twice with culture medium, they were incubated with 50 μM CDDP, with or without 5 mM 4-PBA [26] or 5 mM 3-MA [27]. 8 h later, cells were washed twice with Hanks’ HEPES buffer and observed with a FluoView FV1000 confocal microscope.…”
