Transfection Studies with Colloidal Systems Containing Highly Purified Bipolar Tetraether Lipids fromSulfolobus acidocaldarius

Abstract: Lipid vectors are commonly used to facilitate the transfer of nucleic acids into mammalian cells. In this study, two fractions of tetraether lipids from the archaea Sulfolobus acidocaldarius were extracted and purified using different methods. The purified lipid fractions polar lipid fraction E (PLFE) and hydrolysed glycerol-dialkyl-nonitol tetraether (hGDNT) differ in their structures, charge, size, and miscibility from conventional lipids. Liposomes were prepared by mixing tetraether lipids with cholesterol … Show more

“…In addition, the unique molecular structure of TELs is attributed to the presence of cyclopentane rings, methyl side groups and sugar moieties that create extensive hydrogen bond network. The presence of ether bonds gives rise to molecular stability (Engelhardt et al., 2017 ), in addition to the lacking of double bonds which make TELs an epitome candidate for PDT. One of the elementary factors for tumor progression, for instance, growth, invasion and metastasis, is angiogenesis.…”

“…1,2-distearoyl- sn -glycero-3- phosphatidyl-choline (DSPC) and 1,2-dipalmitoyl-sn-glycero-3-phosphatidyl-choline (DPPC), were a gift from Lipoid GmbH (Ludwigshafen, Germany). A polar lipid fraction containing tetraether lipids caldarchaeol (GDGT ∼10% of the extract) and calditoglycerocaldarchaeol (GDNT ∼90% of the extract) was extracted from the biomass of S. acidocaldarius (SIT Rosenhof GmbH, Heiligenstadt, Germany) as previously mentioned (Engelhardt et al., 2017). (2-hydroxypropyl)-β-cyclodextrin (HPβCD) and all other chemicals and solvents used were of analytical grade and were purchased from Sigma-Aldrich (Taufkirchen, Germany).…”

“…Due to the TELs in the liposomes, a slower release of the drug to the cancer cell could be achieved [131]. In another study, mixing cholesterol and TEL extracted from S. acidocaldarius resulted in a stable vector for gene delivery used for the transfection of mammalian cells [132]. These applications, including the oral delivery of drugs, are summarized in Table 3.…”
