Effect of a one‐step self‐etch adhesive on endogenous dentin matrix metalloproteinases

Abstract: Degradation of the hybrid layer created in dentin by dentin adhesives is caused by enzyme activities present within the dentin matrix that destroy unprotected collagen fibrils. The aim of the present study was to evaluate the effect of a one-step self-etch adhesive system on dentinal matrix metalloproteinases 2 and 4 (MMP-2 and MMP-9, respectively) using in situ zymography and an enzymatic activity assay. The null hypothesis tested was that there are no differences in the activities of dentinal MMPs before and… Show more

“…The tooth-cavity model method described in this study allowed us to quantitate MMP-9 in a clinicallyrelevant sample of dentin isolated from the prepared dentinal cavity walls of a single, extracted tooth. It has been confirmed that the concentrations, the distribution pattern of MMP-9 and the gelatinolytic potential of dentin matrix are variable along different dentin depths 24) . It has also been shown that the predentin and dentinal tubule regions presented higher gelatinolytic activity compared to intertubular dentin.…”

“…Previous studies of endogenous dentin MMPs had been focused on large dentin samples pooled from several extracted teeth. In these studies, the investigators were able to detect gelatinolytic/collagenolytic enzyme activities of endogenous dentin MMPs from large dentin powder samples (between 500 mg and 1 gram) pooled from several extracted teeth and internally quenched fluorescent collagen/gelatin substrate 16,18,[22][23][24] .…”

“…The neutral zinc and calcium dependent matrix metalloproteinases (MMPs) enzymes adjust the physiological and pathological metabolism of collagen-based tissues (Chaussain-Miller, Fioretti, Goldberg, & Menashi, 2006;Visse & Nagase, 2003). When dentin adhesive is applied regardless of self-etch or etch & rinse modes, MMPs can become active due to the calcium ions or acidic components (Apolonio et al, 2017;Mazzoni et al, 2006).…”

“…These MMPs are believed to participate in practically all dental hard tissue pathologies, including dentinal caries [9,10], degradation of the resin-dentin interface of resin composite restorations [11] and dentin erosion [12]. In particular, MMP-2 and MMP-9 are important in caries progression [13,14] and degradation of the resindentin interface [10,[15][16][17].…”
