Development of poly(lactide‐co‐glycolide) nanoparticles functionalized with a mitochondria penetrating peptide

Abstract: The development of mitochondria-targeting cell permeable vectors represents a promising therapeutic approach for several diseases, such as cancer and oxidative pathologies. Nevertheless, access to mitochondria can be difficult. A new hybrid material composed by poly(lactide-co-glycolide) (PLGA) functionalized with a 6-mer mitochondria penetrating peptide (MPP), consisting in alternating arginine and unnatural cyclohexylalanine, was developed. Circular dichroism, FT-IR and DSC studies indicated that the conjuga… Show more

“…3,4,10 NP + can be also obtained by the functionalization of anionic or neutral polymers with cell penetrating peptides, i. e. positively charged oligopeptides composed mainly by Arginine and/or Lysine residues. [11][12][13][14][15][16] In all cases, the polymer is the molecular tool that direct the process of self-assembly while the cationic charge decorates the NP + .…”

“…Given the special properties of these two components, MPPs are skilled at passing across the cell and mitochondria membranes (Jean et al, 2016). Selmin et al developed a mitochondria-targeting hybrid polymer via functionalized PLGA with 6-mer MPP peptide for the first time (Selmin et al, 2017). The 6-mer MPP peptide is composed of alternating arginine and cyclohexylalanine, realizing successful entering the cells and mitochondria.…”
