A novel tomato SUMO E3 ligase, SlSIZ1, confers drought tolerance in transgenic tobacco

Abstract: SUMOylation is an important post-translational modification process that regulates different cellular functions in eukaryotes. SIZ/PIAS-type SAP and Miz1 (SIZ1) proteins exhibit SUMO E3 ligase activity, which modulates SUMOylation. However, SIZ1 in tomato has been rarely investigated. In this study, a tomato SIZ1 gene (SlSIZ1) was isolated and its molecular characteristics and role in tolerance to drought stress are described. SlSIZ1 was upregulated by cold, sodium chloride (NaCl), polyethylene glycol (PEG), h… Show more

“…Conversely, heat‐induced sumoylation of target proteins was observed in several previous studies . In addition, rice E3 SUMO ligases OsSIZ1 and OsSIZ2 were involved in sumoylation of target proteins under heat stress , and transgenic Arabidopsis overexpressing tomato E3 SUMO ligase SISIZ1 also accumulated SUMO conjugates in response to heat stress . There are three types of E3 SUMO ligase in yeast and animal systems: SIZ/PIAS, RanBP2, and polycomb .…”

“…Tobacco seeds were sterilized with 1% sodium hypochlorite for 15 min, washed three times using sterile water, and then sown on 1/2 MS medium. Seven-day-old seedlings were transferred into nutrient soil in a greenhouse at 25/22 • C (day/night) with a 16-h photoperiod and 50-60% relative humidity as described previously (Zhang et al, 2017). Tobacco plants with four leaves were infected.…”

“…umoylation, the covalent attachment of small ubiquitin-like modifier (SUMO) to other proteins, is a post-translational modification that controls protein function, activity, localization, and turnover in eukaryotes 1,2 . In plants, sumoylation is involved in the response to abiotic and biotic stresses, such as cold, salt, and drought stresses, and innate immunity [3][4][5][6][7][8] . Furthermore, sumoylation regulates signaling pathways for plant hormones, including abscisic acid (ABA) and salicylic acid 7,[9][10][11][12] .…”
