is observed in the principal PGB-indicated conditions which share the alteration in IL-6, TNF-α, IL-2 and IL-1β cytokines level. Yet, reports on PGB immunomodulatory and anti-inflammatory effects are a few and focused mainly on in vitro assays, local inflammatory changes within the nervous system, and attenuation of the secretion of only IL-1β and TNF-α cytokines 18-20. In addition, PGB effects on cytokines secretion in immune cells, such as isolated splenocytes and peritoneal macrophages (PMs), and its effect on lymphoid organs, were not examined before. Therefore, there is a need to perform a simultaneous assessment of the effect of PGB on the secretion of the cytokines that were commonly elevated in the aforementioned PGB-indicated conditions (IL-6, TNF-α, IL-1β and IL-2) and to expand the investigation to its effect on lymphoid organs and cytokine secretion in immune cells. As peripheral inflammation and elevated systemic cytokines levels were demonstrated in the above-mentioned PGB-indicated conditions, we investigated in this study the effect of PGB on murine models of peripheral inflammation. In this study, we used for the first time, LPS and ConA-induced murine models of inflammation to examine the effect of PGB on peripheral proinflammatory cytokine secretion (IL-6, TNF-α, IL-1β and IL-2) in vitro and in vivo in BALB/c mice. LPS-model of inflammation has been employed before to study neuroinflammatory conditions, seizure and anxiety disorders in mice 21-25 , while ConA was used to investigate T-cell function in patients with fibromyalgia 26. Additionally, in this study, the effect of PGB on mitogen-induced inflammatory changes in the spleen, as a lymphoid organ, was also examined for the first time. Regarding in vitro investigation, the lack of reports that examined the effect of PGB on the secretion of cytokines in immune cells prompted us to investigate such effects of PGB on basal and mitogen-induced proinflammatory cytokines secretion in splenocytes and peritoneal macrophages (PMs).

Helminths and their products can shape immune responses by modulating immune cells, which are dysfunctional in inflammatory diseases such as asthma. We previously identified SJMHE1, a small molecule peptide from the HSP60 protein of Schistosoma japonicum. SJMHE1 can inhibit delayed‐type hypersensitivity and collagen‐induced arthritis in mice. In the present study, we evaluated this peptide's potential intervention effect and mechanism on ovalbumin‐induced asthma in mice. SJMHE1 treatment suppressed airway inflammation in allergic mice, decreased the infiltrating inflammatory cells in the lungs and bronchoalveolar lavage fluid, modulated the production of pro‐inflammatory and anti‐inflammatory cytokines in the splenocytes and lungs of allergic mice, reduced the percentage of Th2 cells and increased the proportion of Th1 and regulatory T cells (Tregs). At the same time, Foxp3 and T‐bet expression increased, and GATA3 and RORγt decreased in the lungs of allergic mice. We proved that SJMHE1 can interrupt the development of asthma by diminishing airway inflammation in mice. The down‐regulation of Th2 response and the up‐regulation of Th1 and Tregs response may contribute to the protection induced by SJMHE1 in allergic mice. SJMHE1 can serve as a novel therapy for asthma and other allergic or inflammatory diseases.

Background: Helminth infections and their components have been shown to have potential to modulate immunity and attenuate immune response. The objective of this study was to evaluate potential protective effects of Clonorchis sinensis–derived protein (CSp) on ankylosing spondylitis (AS).Methods: Cytotoxicity of CSp at different doses was assessed by MTS and flow cytometry before performing experiments. Peripheral blood mononuclear cells (PBMCs) and Synovial fluid mononuclear cells (SFMCs) were obtained from AS patients. Inflammatory cytokine-producing cells were analyzed using flow cytometry. SKG mice were treated with CSp or vehicle. Inflammation and new bone formation were evaluated using immunohistochemistry, positron emission tomography (PET) and micro–computed tomography (CT).Results: Treatment with CSp resulted in no reduced cell viability of PBMCs or SFMCs. In experiments culturing PBMCs and SFMCs, the frequencies of IFN-g and IL-17A producing cells were significantly reduced after CSp treatment. In the SKG mouse model, CSp treatment significantly suppressed arthritis and enthesitis. Micro-CT analysis of hind paw revealed less new bone formation in CSp-treated mice than in vehicle-treated mice. Conclusions: We provide the first evidence demonstrating that CSp can ameliorate clinical signs and cytokine derrangements in AS. In addition, such CSp treatment could reduce new bone formation of AS.
