Remarkable In Vivo Nonlinear Photoacoustic Imaging Based on Near-Infrared Organic Dyes

Abstract: Two near-infrared dyes featuring good dispersion and light-harvesting property present a remarkable nonlinear photoacoustic response in vitro and in vivo comparing with conventional gold nanorods. This study benefits the fabrication of drug delivery platforms with accurate targeting and control effect under photoacoustic image guidance.

“…Nonlinear PA imaging involves continuous excitation of biological tissue by employing a dual‐pulse laser with a short time delay, which can provide even more significant contrast and resolution than the commonly studied linear PA imaging by reversibly switching the absorption property, or enhancing the thermally related Gruneisen parameter (thermal expansion coefficient) . Some preliminary work about the contrast agents for nonlinear PA imaging has been reported recently . For example, Gao et al designed a contrast agent including two NIR dyes of IR‐820 and IR‐825 for nonlinear PA imaging .…”

“…NIR nanoprobes usually have excellent optical absorption coefficient that can greatly improve the sensitivity of PA imaging, even when facing the challenge of the strong background interference. [ 19–22 ] Simultaneously, the combination of functional aptamer with nanoprobes further facilitates the achievement of non‐invasive diagnosis at molecule level. However, the application of non‐invasive PA imaging in AS disease is still in its infancy.…”

“… 28 , 29 To overcome these problems, a variety of organic photothermal agents have emerged, such as NIR dyes (ICG, IR-820, and IR-825) and RC-BSA. 30 32 However, the low PTC efficiencies of these organic materials are not satisfactory for clinical application. Hence, the rational design and synthesis of effective PTT agents with higher PTC efficiency is in urgent need.…”
