GSK-3β promotes PA-induced apoptosis through changing β-arrestin 2 nucleus location in H9c2 cardiomyocytes

Abstract: Palmitic acid (PA), a type of saturated fatty acids, induces cardiovascular diseases by causing cardiomyocyte apoptosis with unclear mechanisms. Akt participates in PA-induced cardiomyocyte apoptosis. GSK-3β is a substrate of Akt, we investigated its role in PA-induced apoptosis. We reveal that PA inhibits GSK-3β phosphorylation accompanied by inactivation of Akt in H9c2 cardiomyocytes. We also reveal that inhibition the activity of GSK-3β by its inhibitor LiCl or knockdown by siRNA significantly attenuates PA… Show more

“…Accumulating evidence has uncovered the role of β-arrestin2 in apoptosis (23,24) and the dual effect of β-arrestin2 in apoptosis (24)(25)(26)(27). Under different conditions, β-arrestin2 can be pro-apoptotic or anti-apoptotic (28)(29)(30). In the present study, 5-FU, as a core drug for CRC, was selected as a drug-interfering factor.…”

“…The function of GSK-3β in promoting cell apoptosis has been wellelucidated. It not only promoted the signal transduction of apoptosis 46 but also inhibited the expression of antiapoptotic factors. 47 Presumably, the sudden drop in the ovarian index in the ZEA1.5 group 30 was largely due to the apoptosis and atresia of the oocyte, which was in accordance with the results of the increase in GSK-3β.…”

“…C16:0 (palmitic acid), as one of the saturated fatty acids, plays a substantial role in growth and metabolism of body cells. Related studies indicate that the palmitic acid is able to induce cell death and autophagy, and change the phospholipid level in the myocardial cell, thus posing an impact on the functions of mitochondria in cells (Ostrander et al, 2001;Chang et al, 2016). This study shows that the content of C16:0 in yolk of ordinary eggs and native eggs was the highest, without significant difference.…”
