Methyl Protodioscin, a Steroidal Saponin, Inhibits Neointima Formation in Vitro and in Vivo

Abstract: Restenosis (or neointimal hyperplasia) remains a clinical limitation of percutaneous coronary angioplasty. Abnormal proliferation and migration of vascular smooth muscle cells (VSMCs) are known to be involved in the development of restenosis. The present study aimed to investigate the ability and molecular mechanisms of methyl protodioscin (1), a steroidal saponin isolated from the root of Dioscorea nipponica, to inhibit neointimal formation. Our study demonstrated that 1 markedly inhibited the growth and migr… Show more

“…4,5 Although the detailed mechanisms are yet to be determined, it is generally believed that the pathological proliferation and inflammatory responses of vascular smooth muscle cells (VSMCs) can potentially contribute to atherosclerosis and arterial restenosis. 6,7 Therefore, the inhibition of the proliferation of VSMCs and the reduction of the occurrence of restenosis following CEA has become a major research focus.…”

“…Studies have shown that diosgenin and PD can inhibit proliferation, migration, and phenotypic transformation of VSMC and inhibit intima thickening in rat carotid artery balloon injury model through the inhibition of ERK1/2, FOXM1, and ADAM15 expression. 72,73 Therefore, Diosgenin is considered as a promising drug for the treatment of arteriosclerosis and restenosis after PCI.…”

“…Abnormal proliferation and migration of VSMCs is involved in the development of restenosis, and ADAM15 expression in VSMCs plays a critical role in pathological neovascularization and atherosclerosis. Methyl protodioscin inhibited VSMC growth and migration and prevented balloon angioplastyinduced restenosis by downregulating the expression of ADAM15 and its downstream signaling pathways, and thus it may possess some therapeutic potential for the treatment of restenosis (Chung et al, 2016).…”
