Dimethylarginine dimethylaminohydrolase-2 deficiency promotes vascular regeneration and attenuates pathological angiogenesis

Abstract: Ischemia-induced angiogenesis is critical for tissue repair, but aberrant neovascularization in the retina causes severe sight impairment. Nitric oxide (NO) has been implicated in neovascular eye disease because of its pro-angiogenic properties in the retina. Nitric oxide production is inhibited endogenously by asymmetric dimethylarginines (ADMA and L-NMMA) which are metabolized by dimethylarginine dimethylaminohydrolase (DDAH) 1 and 2. The aim of this study was to determine the roles of DDAH1, DDAH2, ADMA and… Show more

“…The laser-induced CNV model was used as previously described [ 15 , 31 , 50 ]. In brief, mice were anaesthetized by intraperitoneal administration of ketamine hydrochloride (100 mg/kg, Pharmacia & Upjohn, Erlangen, Germany) and xylazine (6 mg/kg, Bayer Vital GmbH, Leverkusen, Germany).…”

“…Overexpression of DDAH2 decreased accumulation of ADMA but had no influence on blood pressure [26]. Subsequent examination of DDAH2 -/mice has revealed that ADMA accumulation occurs in the retina following oxygen-induced retinopathy and that this was associated with improved revascularisation [27]. Therefore, evidence suggests that while ADMA can promote vascular dysfunction in many settings, ADMA may also mediate protective effects in the vasculature due to its role in inflammatory cells.…”
