Nifedipine Treatment for Hypertension is Associated with Enhanced Lipolytic Activity and Accelerated Clearance of Postprandial Lipemia

Abstract: Hypertension, advanced age, postprandial hyperlipidemia, and insulin resistance are major risk factors for atherosclerosis. The calcium channel blocker nifedipine is reported to ameliorate insulin resistance possibly by activating PPARγ. This is expected to become accentuated in elderly individuals due to age-related insulin resistance. Insulin resistance modulates lipoprotein metabolism. Therefore, we reasoned that nifedipne offers the potential for improving postprandial lipemia in association with increasin… Show more

“…Forthly, secondary hypertension and dyslipidemia, and genetic familial hypercholesterolemia were not excluded in this study, if any. Finally, the contents of pharmacotherapy for hypertension, which can influence the metabolism of lipids [ 44 , 45 ], the data of renal function and serum uric acid, which contribute to the development of hypertension [ 46 , 47 , 48 , 49 ], and the data of diet were also unavailable in this study. Further studies addressing these limitations are needed to warrant current results.…”

“…Nifedipine was previously shown to decrease total cholesterol, LDL-cholesterol, TG, atherogenic index and elevated HDL level in rats under ethanol and sucrose feeding (dzeufiet). Nifedipine has also significantly decreased fasting TG level and increased HDL-C in the elderly group and was shown to have favorable metabolic effects that are beyond the known enhancement of insulin sensitivity [20].…”

“…To our astonishment, recent work in our lab found that NF caused lipid accumulation and lipotoxicity in the kidneys [22], which showed controversial effects of NF against its other cited benefits [19][20][21]. Recently, it has emerged that the UPR can be directly activated by lipid perturbation, independently of misfolded proteins [23].…”

“…NF lowered the blood pressure and inhibited the development of glomerulosclerosis to the same extent as did moxonidine (a sympatholytic agent) in spontaneously hypertensive rats [19]. In addition, NF was also reported to exhibit many hypolipidemic effects, including enhancing lipolytic activity, accelerating clearance of postprandial lipemia [20], and inhibiting lipid peroxidation [21].…”
