The evolution of tumor metastasis during clonal expansion with alterations in metastasis driver genes

Abstract: Metastasis is a leading cause of cancer-related deaths. Carcinoma generally initiates at a specific organ as a primary tumor, but eventually metastasizes and forms tumor sites in other organs. In this report, we developed a mathematical model of cancer progression with alterations in metastasis-related genes. In cases in which tumor cells acquire metastatic ability through two steps of genetic alterations, we derive formulas for the probability, the expected number, and the distribution of the number of metast… Show more

“…Cancer relapse and metastasis represent major clinical challenges in the treatment of cancer [ 22 , 23 ]. Emerging evidence suggests that tumor cell EMT is an important prerequisite for the generation of CTCs and DTCs, which enables the dissemination of tumor cells into the circulation and surrounding tissues [ 16 , 24 , 25 ].…”

“…Tumor metastasis is still the main cause of cancer-related mortality, so it is important to identify the key molecules in each step of tumor metastasis and develop new strategies for prevention and control of tumor metastasis [ 1 3 ]. A key step in metastasis is that the extravasated carcinoma cells must survive in the foreign microenvironment and ultimately develop into macroscopic metastases.…”

“…Many metastases might be established by a process which lies between these two extreme points, in which a tumor expands due to a balance of consecutive seeding events and subsequent cell divisions. However, previous mathematical models of metastasis have focused almost exclusively on extreme i , single-cell seeding (2832). Although not yet studied in the context of cancer genetics, some mathematical models of consecutive immigration have been applied to other biological systems, in particular island populations (3336).…”
