Bladder Regeneration Using a Smart Acellular Collagen Scaffold with Growth Factors VEGF, FGF2 and HB-EGF

Abstract: Tissue engineering may become an alternative to current bladder augmentation techniques. Large scaffolds are needed for clinically significant augmentation, but can result in fibrosis and graft shrinkage. The purpose of this study was to investigate whether smart acellular collagen-heparin scaffolds with growth factors (GFs) VEGF, FGF2, and HB-EGF enhance bladder tissue regeneration and bladder capacity in a large animal model of diseased bladder. Scaffolds of bovine type I collagen with heparin and VEGF, FGF2… Show more

“…9,[31][32][33] There are many bioactive factors which can be added to the scaffold, for example BMP, 1,2,[14][15][16]34 other growth factors including VEGF, FGF-2, HB-EGF, PDGF-BB, etc. 33,[35][36][37][38] and drugs 23 such as icariin, 30 licochalcone, 39 alendronate 40 and so on, among which BMP is the most frequently used in bone materials. BMP has been proved to have a high efficacy of osteoinductivity; however, it is seldom used to promote bone formation alone by the reason of being easily diluted and degraded.…”

“…All of these methods are applicable for growth factors modified on a collagen-based scaffold. For example, VEGF, FGF-2 and heparin-binding epidermal growth factor (HB-EGF) were loaded on collagen scaffold by physical adsorption through soaking scaffolds in phosphate buffered saline (PBS) containing growth factors [93]. Li et al [94] fused stromal cell-derived factor-1α (SDF-1α) with a unique peptide of the collagen binding domain (CBD), so the CBD-SDF-1α can specifically bind to the collagen scaffold.…”
