Proteoliposomes with the ability to transport Ca2+ into the vesicles and hydrolyze phosphosubstrates on their surface

Abstract: We describe the production of stable DPPC and DPPC:DPPS-proteoliposomes harboring annexin V (AnxA5) and tissue-nonspecific alkaline phosphatase (TNAP) and their use to investigate whether the presence of AnxA5 impacts the kinetic parameters for hydrolysis of TNAP substrates at physiological pH. The best catalytic efficiency was achieved in DPPS 10%-proteoliposomes (molar ratio), conditions that also increased the specificity of TNAP hydrolysis of PPi. Melting behavior of liposomes and proteoliposomes was analy… Show more

“…The first step is the deposition of an amorphous mineral complex, i.e. the nucleation core (NC), which crystalizes to form HA inside MVs [58]. Concomitantly, MVs bleb out from cell membranes, bind collagen fibrils through specific molecular interactions and decompose to release their HA crystals, enabling HA propagation onto the collagenous ECM.…”

“…Biophysical and biochemical characterization of biomimetic models Biophysical tools applied to biomineralization studies A large number of studies have been carried out using proteoliposomes as chondrocyte-and osteoblast-derived MVs biomimetics (Andrade et al 2016;Bolean et al 2010Bolean et al , 2011Bolean et al , 2015Bolean et al , 2017Ciancaglini et al 2010;Favarin et al 2017;Simao et al 2010aSimao et al , b, 2013.…”

“…Proteoliposomes can be used to study how the protein-lipid interactions specifically affect the surface of the membrane model. The scientific community has highlighted this approach as an excellent tool to understand biochemical and biophysical phenomena and to evaluate biotechnological applications (Camolezi et al 2002;Ierardi et al 2002;Daghastanli et al 2004;De Lima Santos et al 2005;Rigos et al 2008;Santos et al 2009;Bolean et al 2010Bolean et al , 2011Bolean et al , 2015Simão et al 2010aSimão et al , 2010bBarbosa et al 2011;Yoneda et al 2013Yoneda et al , 2014Simão et al 2015;Dong et al 2016;Elkhodiry et al 2016). …”
