2,3-Dimethoxy-2′-hydroxychalcone ameliorates TNF-α-induced ICAM-1 expression and subsequent monocyte adhesiveness via NF-kappaB inhibition and HO-1 induction in HaCaT cells

Abstract: Up-regulation of adhesion molecules plays an important role in the infiltration of leukocytes into the skin during the development of various inflammatory skin diseases, such as atopic dermatitis. In this study, we investigated the modulatory effects of 2,3-dimethoxy-2′-hydroxychalcone (DMHC) on tumor necrosis factor (TNF)-α-induced intercellular adhesion molecule-1 (ICAM-1) expression and monocyte adhesiveness, as well as the molecular mechanisms underlying its action in the HaCaT human keratinocyte cell line… Show more

“…Against the inflammatory cytokines (i.e., tumor necrosis factor‐alpha [TNF‐α] and interferon‐gamma), ICAM‐1 expression levels can be increased in the keratinocytes of psoriatic lesions (Kim et al, ). The upregulation of ICAM‐1 expression implies the progress of inflammatory disorders, and ICAM‐1 may act as a critical role in the interactions between keratinocytes and leukocytes (Kim et al, ; Matsunaga, Katayama, Yokozeki, & Nishioka, ). As shown in Figure , ICAM‐1 expression level in IMQ group was significantly higher than that of control group.…”

“…Of note, HO-1 expression has been reported to exert an anti-inflammatory response in keratinocytes, and attenuates AD-like lesions via its protective effects against inflammatory skin diseases (7,9,15). HO-1 expression is regulated by inflammation-associated cytokines and chemokines associated with the development of AD (15).…”

“…ICAM-1 (7)(8)(9). Additionally, TNF-α-induced expression of inflammatory mediators within keratinocytes simultaneously activates inflammatory signaling pathways, nuclear factor-κB (NF-κB) and mitogen-activated protein kinases (MAPKs), resulting in aggravated inflammation (10).…”

“…Samples were subjected to 10% SDS-polyacrylamide gel electrophoresis and subsequently transferred onto nitrocellulose membranes, detected by enhanced chemiluminescence according to the manufacturer's instructions (Amersham, Buckinghamshire, UK), and analyzed using a Molecular Imager ChemiDoc XRS System (Bio-Rad, Hercules, CA) as described elsewhere (Eom et al 2015;Kim et al 2016). The following antibodies were used for Western blotting: anti-caspase-3, anti-calpain, anti-Bax, anti-Bcl-2, anti-iNOS, antinNOS, anti-Nrf2, anti HO-1, and anti-GCLC.…”
