Endothelial Cells Control Pancreatic Cell Fate at Defined Stages through EGFL7 Signaling

Abstract: SummaryAlthough endothelial cells have been shown to affect mouse pancreatic development, their precise function in human development remains unclear. Using a coculture system containing human embryonic stem cell (hESC)-derived progenitors and endothelial cells, we found that endothelial cells play a stage-dependent role in pancreatic development, in which they maintain pancreatic progenitor (PP) self-renewal and impair further differentiation into hormone-expressing cells. The mechanistic studies suggest that… Show more

“…The vasculature can secrete specific factors at the sites of tissue injury that can influence tissue regeneration, as seen in the lung (66) and liver (67). This effect has also been noted in various tissues, including: pancreatic islet cells (68), adipose stem cells (69), bone (70, 71) and cardiac endothelium (72). Beginning in 2000, Rafii and other groups showed that it was possible to produce organ-specific endothelial cells (7376).…”

“…Angiocrine signals from ECs also guide the stage-specific differentiation of pluripotent stem cells into islet cells. For example, EC-derived EGF-like 7 (EGFL7) specifies the fate and proliferation of pancreas and duodenum homeobox (Pdx)1-positive islet-cell progenitors 100 . Therefore, identification of the phenotypic and functional attributes of islet-specific ECs could unravel the angiocrine heterogeneity within pancreatic tissues and lead to new therapeutic strategies for islet-cell production.…”

“…41,42 Once they reached B80-90% confluency they were treated for one day with 3 mm CHIR99021 (StemRD) and 100 ng mL À1 activin A (R&D systems) in RPMI medium (Cellgro) supplemented with 2 mM GlutaMAX (Gibco) and 100 U mL À1 Pen/Strep (Gibco). For the following two days the cells were treated with 100 ng mL À1 activin A in RPMI medium supplemented with 0.2% FBS, 2 mM GlutaMAX and 100 U mL À1 Pen/Strep.…”
