Drug Resistance

Abstract: Drug resistance is a fundamental problem in the treatment of cancer since cancer that becomes resistant to the available drugs may leave the patient with no therapeutic alternatives. In this chapter, we consider the dynamics of drug resistance in blood cancer and the related issue of the dynamics of cancer stem cells. After describing the main types of chemotherapeutic agents available for cancer treatment, we review the different mechanisms of drug resistance development. Various mathematical models of drug r… Show more

“…Overcoming the emergence of drug-resistance or combatting persistence cells (persisters) is a major challenge in DD. 201 204 Kohanski et al discussed the different drug resistant mechanisms from a biological network viewpoint. 205 Liu et al made an in silico investigation to study intrinsic and induced drug resistance mechanism in solid tumor by bridging the gap between cell and tissues.…”

“…Many "biological drugs" based upon a strong rationale related to the understanding of well-defined pathological mechanisms are now available for clinical oncologists [18,19]. Promising or even excellent results have been obtained in some types of tumors.…”

“…Promising or even excellent results have been obtained in some types of tumors. However, used individually or in combination with a more conventional treatment, such approaches so far have not substantially improved the prognosis of most types of advanced-stage tumors [18,19].…”

“…Historically, hematological neoplasms have been treated with conventional chemotherapy drugs and, more recently, with combined therapies. However, despite the broad number of therapeutic options available, many treatments eventually generate partial or total resistance in many patients, leading to drug insensitivity in cancer cells and, consequently, treatment ineffectiveness [ 4 ]. In an attempt to elucidate the cellular and molecular mechanisms underlying the process of resistance to therapy, numerous investigations have identified potential therapeutic targets in multiple cellular functions [ 5 ].…”
