Ano1 is a better marker than c-Kit for transcript analysis of single interstitial cells of Cajal in culture

Abstract: Abstract:The interstitial cells of Cajal (ICC) drive the slow wave-associated contractions in the small intestine. A commonly used marker for these cells is c-Kit, but another marker named Ano1 was recently described. This study uses single-cell RT-PCR, qPCR and immunohistochemistry to determine if Ano1 could be reliably used as a molecular marker for ICC in single-cell mRNA analysis. Here, we report on the relationship between the expression of c-Kit and Ano1 in single ICC in culture. We observed that Ano1 is… Show more

“…Another marker, considered by some authors even better than the c-Kit for the ICC identification (29), was recently found; the anoctamin 1 (ANO1). It is a Ca 2+ -activated chloride channel necessary for slow wave generation and devoid of any effect on ICC differentiation (30).…”

“…C-kit labeling indirectly reflects the quantity and density of ICC [47,48]. It can not only maintain the phenotype of ICC, but also determine the differentiation, development, and rhythmic activity of ICC [49,50]. Our results indicate that YTC increased the expression level of C-kit and SCF, which protect the expression of ICC.…”

“…Furthermore, ICCs were identified by immunofluorescent double staining. ANO1 is a new and highly selective molecular marker for ICCs [ 34 ]. The cultured cells were positive for both c-kit and ANO1, indicating that they were ICCs.…”
