Background Finding new effective substances to develop drugs against cancer or to prevent it remains a big challenge facing scientists and clinicians. Objectives In such an issue, we tested the anti-tumoral effect of Scorzonera undulata extracts on MCF7 cells. Materials and Methods Methanolic extracts of S. undulata roots (RSU) and aerial parts (ASU) from the Ha’il region were analyzed by HPLC. Their DPPH scavenging, ferric-reducing antioxidant power (FRAP) and total antioxidant potentials were evaluated. The MTT test was used to determine their cytotoxic effects on MCF7 cells using different concentrations, and their potential to induce apoptosis was determined by measuring the mitochondrial transmembrane loss of potential by flow cytometry using the JC-1 fluorescent marker. Results S. undulata was found to be rich in flavonoids, tannins, and polyphenols. ASU contains great amounts of apigenin and gallic acid, whereas RSU contains luteolin and chlorogenic acid as major components. The potential to scavenge DPPH and the total antioxidant activity of ASU were twofold greater than those of RSU. MCF7 cell viability was reduced by 50% at concentrations of 4.22 ± 0.06 and 5.89 ± 0.08 mg.mL−1, respectively, for ASU and RSU at 24 h. In addition, S. undulata extracts induced MCF7 cells’ death through cell lysis and apoptosis. Conclusion As an edible vegetable, S. undulata is worth further exploration in alternative and complementary medication as it possesses significant antioxidant and anticancer potentials.

Leaves of Scorzonera incisa DC. (Asteraceae) are eaten raw against diabetes and headache. Evaluation of medicinal, edible plants using profiling methods has become a common phenomenon. Petroleum ether (PE), ethyl acetate (EA), and n‐butanol (BU) fractions were prepared from ethanol extract of the aerial parts of the plant. Isolation by HPLC and phenolic profiling by validated LC‐MS/MS method with 53 compounds were conducted on the EA fraction. GC‐MS method was used to profile the PE fraction. In‐vitro inhibitory activities of all fractions against α‐amylase, α‐glucosidase, AChE, BChE, tyrosinase and urease enzymes were measured. Isoorientin, vitexin, cynaroside and chlorogenic acid were isolated and characterized from the EA fraction as major compounds. LC‐MS/MS analysis quantified 36 compounds in the EA fraction. 20 compounds were tentatively determined in the PE fraction. Compounds with the same molecular and fragment ions, a low percentage of identified compounds, and very close polarities were revealed to be some limits of the nature for LC‐MS/MS profiling. Particularly, the presence and amount of substances other than the standard compounds used in the profiling methods and possible misdetections should not be neglected in evaluating medicinal plants and establishing a correlation between compounds and bioactivity.

Callus Culture of Scorzonera radiata as a New, Highly Productive and Stable Source of Caffeoylquinic Acids

During our ongoing efforts to investigate biotechnological sources of caffeoylquinic acid (CQA) metabolites, we discovered the plant Scorzonera radiata Fisch. (Asteraceae), which is able to produce callus cultures with high yield and extremely high stability. An actively growing callus line, designated as Sr-L1, retained the ability to produce 11 CQAs during long-term cultivation (more than 20 years). A total of 29 polyphenolic compounds were identified in the leaves and Sr-L1 callus culture of S. radiata, including CQAs, lignol derivatives, flavonoids, and dihydrostilbenes. The composition of CQAs in the Sr-L1 culture was identical to that in the S. radiata leaves. Sr-L1 calli did not produce flavonoids and dihydrostilbenes, but produced lignol derivatives, which were absent in leaves. The HPLC-UV-HRMS determination showed the presence of monoacyl derivatives of CQAs such as 5-CQA, 4-CQA, cis-5-CQA, and 5-O-p-coumaroylquinic acid in the Sr-L1 culture. Among diacyl derivatives, 3,4-diCQA, 3,5-diCQA, cis-3,5-diCQA, 4,5-diCQA, 3-O-p-coumaroyl-5-O-CQA, and 3-O-caffeoyl-5-O-p-coumaroylquinic acid were found. The content of 5-CQA reached 7.54 mg/g dry weight and the content of 3,5-diCQA was as high as 18.52 mg/g dry weight. 3,5-diCQA has been reported to be of high nutritional and pharmacological value, as it alleviates inflammatory pain, reverses memory impairment by preventing neuronal apoptosis, and counteracts excessive adipose tissue expansion, serving as an attractive treatment option for obesity. The high content of 3,5-diCQA and the exceptional stability of biosynthesis make callus cultures of S. radiata a promising source for the development of drugs and nutraceuticals.
