Transcriptional and epigenetic basis for restoration of G6PD enzymatic activity in human G6PD-deficient cells

Abstract: Key Points Increase in HDAC binding is required for HDAC inhibitors to enhance gene transcription. G6PD deficiency in erythroid precursors can be restored by HDAC inhibitor-mediated increased transcription of the variant gene.

“…For example, an HDACi, givinostat, showed a therapeutic benefit for patients with juvenile idiopathic arthritis by reducing the production and release of several proinflammatory cytokines [38]. HDACi is also known to restore the expression of defective enzymes [39], channels [40], and growth factors [41]. In these reports, HDACi enhanced the transcription of the target gene beyond the basal levels, and restored the amount of protein by histone hyperacetylation of the promoter region [39].…”

Similar to those previous reports, in the present study, TSA restored the EMT-induced reduction of SFTPC mRNA expression beyond the basal level by the hyperacetylation of histone H4 in the SFTPC promoter region in vitro.…”

“…[46][47][48] Because adipose tissue simultaneously faces those signaling pathways in obesity, further studies are needed to precisely delineate the signaling pathways leading to upregulation of G6PD in obesity. In addition, another open question is what molecular events are responsible for dual, opposing effects of G6PD deficiency on ROS balance in different cell types.…”

“…Detection of hemoglobinuria by urine dipsticks is important. In fact, the presence of hemoglobinuria in spite of the presence of a level of hemoglobin of 9 g/dL may indicate immediate blood transfusion of packed RBCs because hemoglobinuria may indicate persistent brisk hemolysis, which if left will cause blockage of renal tubules leading to acute renal failure [9][10][11][12][13][14][15].…”
