Association of interleukin-10 gene promoter polymorphisms with susceptibility to acute pyelonephritis in children

Abstract: Interleukin-10 (IL-10) is a potent inhibitor of leukocyte chemotaxis, bacterial killing in phagocytes and synthesis of pro-inflammatory cytokines and chemokines, and recent studies have suggested an important role for this immunoregulatory cytokine in the pathogenesis of urinary tract infections (UTIs). Therefore, the gene encoding IL-10 (IL10) is an attractive candidate for association studies attempting to identify susceptibility genes conferring risk of UTIs. In this case-control study, we aimed to investig… Show more

“…Also in UTI genetic factors likely play a role, as illustrated by the finding that positive family history is a risk factor for recurrent UTI [ 23 ] and reviewed in [ 1 ]. Studies evaluating the role of genetic factors in UTI were mainly performed in children [ 24 29 ]. No studies have investigated the contribution of host genetic factors to the development of UTI complicated by bacteremia.…”

“…IL-10 is produced by T-lymphocytes, and is an important anti-inflammatory and immune-suppressive cytokine that regulates angiogenesis in many inflammation-related diseases (Bantis et al, 2008;Javor et al, 2014;Peng et al, 2015;Yu et al, 2015). The IL-10 gene is a candidate gene in the pathophysiological mechanism of auto-immune/inflammatory disease, as it can regulate both the cellular and humoral immunity.…”

“…Mechanisms of innate immune activation and essential effector functions in APN have been mapped using cellular infection technology, animal models and clinical studies 4 , 10 , 11 , 14 , 16 , 18 , 19 , 32 , 75 80 . Key regulators of UTI severity have been evaluated using gene knockout technology and their relevance to human disease has been verified in clinical studies 10 , 11 , 29 , 78 .…”
