Transcriptome analysis of Inner Mongolia Cashmere goat and Dazu black goat generated 38,772,947 and 38,771,668 clean pair end reads, respectively, which were assembled into 72,422 and 80,069 unigenes by Trinity, respectively. For Inner Mongolia Cashmere goat, 26,051 and 10,100 unigenes were assigned to gene ontology (GO) categories and clusters of orthologous groups, respectively. A total of 32,772 unigenes can comment to SWISS-Prot database, and the Kyoto Encyclopedia of Genes and Genomes Pathway database (KEGG) mapped 24,420 unigenes. While annotating the unigenes about Dazu black goats, we found 29,444(45.42%), 11,271 (38.28%), 36,910(56.94%) and 27,766 (42.83%) unigenes were assigned to GO database, COG database, SWISS-Prot database and KEGG database, respectively. In addition, we performed the bioinformatics analysis of gene expression profiling aimed at the ovarian transcriptome difference between Inner Mongolia Cashmere goat and Dazu black goat. We obtained a sequencing depth of over 5.5 million and 5.8 million tags. There were 1,133 DEGs between two species, of which 632 genes upregulated in the Dazu black goat and 501 genes downregulated compared with which in Inner Mongolia Cashmere goat. By annotating the 1,133 DEGs into KEGG database, we found 525 DEGs. And there were 68 DEGs annotated in metabolic pathways, 31 DEGs annotated in ribosome, 28 DEGs annotated in focal adhesion, 27 DEGs annotated in phagosome, 26 DEGs annotated in pathways in cancer, 25 DEGs annotated in ECM-receptor interaction, 23 DEGs annotated in protein digestion and absorption, 20 DEGs annotated in oxidative phosphorylation, 17 DEGs annotated in lysosome, and 16 DEGs annotated in cell adhesion molecules.

Sericin is a water-soluble component of silk and has been used as a biomaterial due to its antibacterial and ultraviolet radiation-resistant properties. This study was designed to evaluate the effect of sericin supplementation in a maturation medium on the meiotic competence and fertilisability of sheep oocytes. Cumulus-oocyte complexes (COCs) were cultured in TCM199 supplemented with sericin at various concentrations of 0 (control), 0.1, 0.25 and 0.5%, either with or without bovine serum albumin (BSA). When the COCs were matured without BSA, the supplementation of 0.1% sericin significantly increased the rates of maturation to metaphase II and the total fertilisation of oocytes compared with the other concentrations of sericin. When the COCs were matured with BSA, the beneficial effects of 0.1% sericin supplementation on the maturation and fertilisation of oocytes were not observed. Our findings indicate that supplementation with 0.1% sericin during maturation culture may improve the nuclear maturation and fertilisability of sheep oocytes. Moreover, it may be possible to replace BSA with sericin in chemically defined media without the risk of disease transmission.

In vitro embryo production (IVEP) contributes to the quantitative and qualitative aspects of animal reproduction. Nevertheless, inherent technical factors such as oxidative stress can negatively influence the result and this can impair cell metabolism, thus decreasing the rates of in vitro development, and necessitating the supplementation of culture medium with antioxidants. In this context, compounds of natural origin with this property have been highlighted because of the positive results obtained at different stages of IVEP. Thus, this review aims to present the results obtained by using natural antioxidants to minimize the effects of oxidative stress on gametes and embryos. A variety of natural isolated substances and mixtures (essential oils and extracts) have been studied for supplementation of IVEP media, at stages of in vitro maturation, sperm capacitation, in vitro fertilization, and in vitro development of embryos in different mammalian species. Generally, beneficial effects are observed according to the concentration used, thus demonstrating the potential of several natural antioxidants. Therefore, the main challenges in using these compounds as antioxidants during IVEP include proving their efficiency against free radicals and determining the best concentration at each stage. In addition, understanding the mechanisms of action of such antioxidants is crucial to establishing their use in IVEP biotechnology. Key words: Culture medium. Natural bioactive. Oxidative stress. Reproduction. ResumoA produção in vitro de embriões (PIVE) contribui para os aspectos quantitativos e qualitativos da reprodução animal. Contudo, fatores inerentes da técnica, como o estresse oxidativo, podem influenciar negativamente o resultado e isso pode prejudicar o metabolismo celular, diminuindo assim as taxas de desenvolvimento in vitro, e exigindo a suplementação do meio de cultivo com antioxidantes. Neste contexto, os compostos de origem natural com essa propriedade têm se destacado devido aos resultados positivos obtidos em diferentes estágios da PIVE. Assim, esta revisão pretende apresentar os resultados obtidos usando antioxidantes naturais para minimizar os efeitos do estresse oxidativo em gametas e embriões.
