The effect of platelet‐derived microparticles in stored apheresis platelet concentrates on polymorphonuclear leucocyte respiratory burst

Abstract: The platelet-derived microparticles, which carried the sCD40L, accumulated in the apheresis platelet concentrates during the 5 days of storage. They primed the fMLP-activated PMN respiration burst, which might be relative to TRALI.

“…[58][59][60] Soluble CD40L activates macrophages and elicits the production and release of multiple pro-inflammatory cytokines. Increased sCD40L in stored platelets has been associated with transfusion reactions [61] and increased respiratory burst in polymorphonuclear leukocytes (PMNs) [62]. In animal models, acute lung injury caused by LPS or reactive oxygen species (ROS) is attenuated by inhibition of the CD40-CD40L system [56,63].…”

“…Our recent study demonstrated that PLT‐derived microparticles (PMPs) carried concentrated CD40L signal. These microparticles effectively prime formyl‐Met‐Leu‐Phe (fMLP)‐activated PMN respiration burst . However, the effects of PMPs from PLT concentrates on endothelial cells are unknown.…”

“…In addition to RBCs, the platelet-derived MPs increase during the storage period, prime the fMLP-activated PMN respiration burst, which may induce TRALI [59]. Moreover, monocyte-derived MPs up regulate the level of pro-inflammatory factors in lung epithelial cells, primarily through activating NF-κB and PPAR-γ dependent pathways [60].…”
