Bidirectional fluxes of spermine across the mitochondrial membrane

Abstract: The polyamine spermine is transported into the mitochondrial matrix by an electrophoretic mechanism having as driving force the negative electrical membrane potential (DW). The presence of phosphate increases spermine uptake by reducing DpH and enhancing DW. The transport system is a specific uniporter constituted by a protein channel exhibiting two asymmetric energy barriers with the spermine binding site located in the energy well between the two barriers. Although spermine transport is electrophoretic in or… Show more

“…On the other hand, conjunction with PAs may result in agents capable of selectively exploiting the highly active PA-transporters (PAT) in cancer cells (27). In addition, the PA backbone would recognize the ionic surface of mitochondria and penetrate these organelles (28). Both properties render PA–CAM conjugates promising anticancer agents.…”

“…Also the effects of polyamines on the conductance of larger pores like OmpF [ 26 , 29 ] and the translocase of the outer chloroplast membrane Toc75 have been tested [ 32 ]. A potential physiological role of spermine flux across the mitochondrial membranes is summarized in [ 33 ].…”

“…Polyamines interact with mitochondrial membranes at specific sites involved in their electrophoretic transport in mitochondria (Grancara et al 2014). In particular, spermine can also flow from organelles by promoting the crossmembrane cycling which regulates the induction of MPT (Agostinelli et al 2010).…”
