Phospholipase gene expression during Paracoccidioides brasiliensis morphological transition and infection

Abstract: Phospholipase is an important virulence factor for pathogenic fungi. In this study, we demonstrate the following: (i) the Paracoccidioides brasiliensis pld gene is preferentially expressed in mycelium cells, (ii) the plb1 gene is mostly up-regulated by infection after 6 h of co-infection of MH-S cells or during BALB/c mice lung infection, (iii) during lung infection, plb1, plc and pld gene expression are significantly increased 6-48 h post-infection compared to 56 days after infection, strongly suggesting that… Show more

“…Phosphatidic acid and diacylglycerol produced by a phospholipase D1 participates in the morphological transition of C. albicans ( Hube et al, 2001 ). A similar finding was seen in P. brasiliensis in which up-regulation of phospholipase was found in M-Y transition ( Soares et al, 2013 ).…”

“…Virulence factors are essential for establishing an invasive fungal infection, including dimorphism, host cell adhesion, cell wall composition, and hydrolytic enzyme production. (16,17,18,19,20,21,40) Another aspect of this disease is pulmonary fibrosis, which is char- acterised by excessive deposition of collagen and extracellular matrix components that results in pathological remodeling of the pulmonary architecture. (40) In this study, the expression of six genes in the Y and M forms of Pb18 and Pb01 after infection of two different cell types was determined using qPCR.…”

“…Enrichment analyses of Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways ( ) were used to functionally categorize the regulated proteins and interactions between proteins were studied on the STRING v.11 server [ 26 ], available at . Proteins associated with virulence, stress response, and pathogenesis of paracoccidioidomycosis were retrieved from indexed publications [ 9 , 27 , 28 , 29 , 30 , 31 , 32 , 33 ] and searched into the proteomic data generated in this study. Heat maps were constructed using the GraphPad Prism 8.4.2 software.…”
