Reduced expression of NGEP is associated with high-grade prostate cancers: a tissue microarray analysis

Abstract: New gene expressed in prostate (NGEP) is a newly diagnosed prostate-specific gene that is expressed only in normal prostate and prostate cancer cells. Discovery of tissue-specific markers may promote the development of novel targets for immunotherapy of prostate cancer. In the present study, the staining pattern and clinical significance of NGEP were evaluated in a series of prostate tissues composed of 123 prostate cancer, 19 high-grade prostatic intraepithelial neoplasia and 44 samples of benign prostate tis… Show more

“…Immunohistochemical staining was performed on TMA slides (Superfrost plus, Thermo Scientific, Germany) prepared from formalin-fixed paraffin-embedded tissues (FFPE) using a protocol as previously described (Mohsenzadegan et al, 2013;Taeb et al, 2014). Briefly, after deparaffinizationin xylene, tissues were rehydrated by immersion in decreasing grades of ethanol and endogenous peroxidase activity was blocked by immersing the tissues for 20 min in methanol containing 0.3% hydrogen peroxide and retrieved by boiling in a microwave for 20 minutes with sodium citrate buffer (pH 6.0).…”

“…Immunohistochemistry was performed on the TMA slides (Superfrost plus, Thermo Scientific, Germany) with a standard chain polymer-conjugated (Envision) technique as described previously (Madjd et al, 2011;Mohsenzadegan et al, 2013) DOI:http://dx.doi.org/10.7314/APJCP.2014.15.19.8161 Expression of CD133 and Nestin Cancer Stem Cell Markers in Skin Cancers antibodies. The slides were deparafinized in 60ÂșC for 20 minutes and cleared in xylene and were then rehydrated in ethanol.…”

“…Skin cancer tissue microarrays were constructed as described previously (Kononen et al, 1998;Mehrazma et al, 2013;Mohsenzadegan et al, 2013). All Hematoxylin and Eosin (H and E)-stained slides were reviewed by a pathologist (AR) to determine the best area for preparing the TMA of each specimen.…”

“…The paraffin-embedded biopsy specimens were applied to construct tissue microarray blocks as described previously (Mehrazma et al, 2013;Mohsenzadegan et al, 2013;Taeb et al, 2014). Briefly, the regions of interest in paraffin-embedded tissues were identified and marked out using 3-”m H&E slides to obtain the cores.…”
