A Novel Role for Tm7sf2 Gene in Regulating TNFα Expression

Abstract: We have explored the role of Tm7sf2 gene, which codifies for 3β-hydroxysterol Δ14-reductase, an endoplasmic reticulum resident protein, in the sensitivity to endoplasmic reticulum stress and in the resulting inflammatory response. We used mouse embryonic fibroblasts, derived from Tm7sf2+/+ and Tm7sf2−/− mice, to determine the in vitro effects of thapsigargin on NF-κB activation. Our results show that the Tm7sf2 gene controls the launch of the unfolded protein response and presides an anti-inflammatory loop thu… Show more

“…Can other factors affect the impairment in proliferation observed in absence of Tm7sf2 gene? Recently, it has been determined that Tm7sf2 KO mice present an altered ER stress response following different stress inducing treatments 27 and liver regeneration represents a strong stress condition in ER. Moreover, it has been demonstrated that lipids accumulated in hepatocytes after PH can trigger ER stress, which in turn may result in a delay in DNA replication and impairment of liver regeneration.…”

“…Transmembrane 7 superfamily member 2 (Tm7sf2) functions just downstream of Sqle in the cholesterol biosynthesis pathway, and its function is linked to liver X receptor (LXR) and NFκβ signaling, suggesting roles in both lipid homeostasis and inflammation (Bellezza et al 2013).…”
